There are many drying methods, such as sunlight drying, boiling drying, drying, spray drying and vacuum drying, etc., but common drying methods are usually carried out at temperatures above 0°C. Dried products usually have the problems of volume reduction and hard texture. Most of the volatile components will be lost.
Some heat-sensitive substances will denature and inactivate, and some will even oxidize. Therefore, compared with before drying, the properties of the dried product are quite different. The freeze-drying method is basically carried out below 0°C, that is, the product is frozen, and the product is raised to a temperature above 0°C only when the residual moisture of the product is reduced later but usually does not exceed 0°C. 40°C.
Under vacuum conditions, when water vapor directly sublimates, the drug remains on the ice shelf when frozen, forming a spongy loose porous structure, so its volume is almost unchanged after drying. Before using it again, just add water for injection and it will dissolve immediately. Compared with traditional methods, freeze dryers have the following advantages:
*Many heat-sensitive substances will not be denatured or inactivated.
*When drying at low temperature, some volatile components in the material lose little.
*During the freeze-drying process, the growth of microorganisms and the action of enzymes cannot be carried out, so the original characteristics can be maintained.
*Because it is dried in a frozen state, there is almost no change in volume, the original structure is maintained, and concentration does not occur.
*Because the water in the material exists in the form of ice crystals after pre-freezing, the dissolved substances of the inorganic salt initially dissolved in the water are evenly distributed in the material. During the sublimation process, the dissolved substances dissolved in the water will precipitate out, avoiding the surface hardening phenomenon caused by the precipitation of inorganic salts carried by the water inside the material on the surface in the general drying method.
*The dry substance is loose, porous and sponge-like. After adding water, it dissolves quickly and completely, and restores its original performance almost immediately.
*Because drying is carried out under vacuum, there is little oxygen, so it can protect some easily oxidized substances.
*Drying can remove 95% to more than 99% of moisture, so the dried product can be stored for a long time without deterioration.
*Because the material is frozen and the temperature is extremely low, the temperature of the heating source is not high, and the normal temperature or low temperature heater can meet the requirements. If the freezer compartment and the drying compartment are separated, the drying compartment does not need to be insulated, and there will not be a large amount of heat loss, so the use of heat energy is very economical.
Since there is no perfect technology, the main disadvantage of vacuum freeze-drying technology is high cost. Due to the need for vacuum and low temperature conditions, the vacuum freeze dryer should be equipped with a vacuum system and a low temperature system, so the investment cost and operating cost are high.
